Rough Eye Quantification

Rough eye quantification is a phenotypic analysis method that measures the severity of disrupted eye morphology, commonly in developing organisms such as Drosophila, to assess genetic or environmental effects on retinal development. The method compares standardized eye images or specimens for features such as ommatidial disorganization, surface irregularity, and overall structural defects, converting visible abnormalities into qualitative scores or quantitative measurements. Researchers use this assay to evaluate gene function, signaling pathways, and genetic interactions that shape eye formation. Because it provides a rapid, reproducible readout of developmental defects, rough eye quantification supports screening and mechanistic studies in developmental biology.

This video demonstrates the phenol red thread test to determine tear volume in an autoimmune dry eye rat model. The pH-sensitive acidic phenol red thread changes color when it comes in contact with the alkaline tears. The shorter length of color change in the phenol red thread corresponds to reduced tear volume.

This work has discussed the protocol for designing and developing an E-Eye-Enable POCT device to detect Fe, Cr, As, and F in environmental samples, biological samples, and food and beverages. Nearly 2000 samples have been tested, and the results accord with the gold-standard technique.

Preparation of Adult Drosophila Eyes for Thin Sectioning and Microscopic Analysis

A standard approach to prepare adult Drosophila eyes for semi-thin sectioning and light microscopic analysis is presented here. The protocol can be used for gross morphological analysis of eye defects, or with the indicated adjustments can be used to determine genetic requirements of genes in specific cell types of the eye (e.g. clonal analysis of photoreceptors) or for electron microscopic analysis.

Isolation of Retinal Stem Cells from the Mouse Eye

In this video, we will demonstrate how to isolate retinal stem cells from the ciliary epithelium of the mouse eye and grow them in culture to form clonal retinal spheres. The spheres that are isolated possess the cardinal properties of stem cells: self-renewal and multipotentiality.
