Protein Film Hydration

Protein film hydration is the uptake and incorporation of water into a dry protein-based film, a process that strongly influences the film’s structure and performance in engineering applications. Water diffuses through the matrix and forms hydrogen bonds with polar amino acid groups, disrupting some protein-protein interactions, increasing chain mobility, and potentially causing swelling or plasticization. These changes can alter tensile strength, flexibility, permeability, degradation, and adhesion. Studying hydration helps engineers design protein coatings, biodegradable packaging, and biomaterial films with controlled mechanical and barrier properties under specific environmental conditions.

Here, we describe a technique, protein film infrared electrochemistry, which allows immobilized redox proteins to be studied spectroscopically under direct electrochemical control at a carbon electrode. Infrared spectra of a single protein sample can be recorded at a range of applied potentials and under a variety of solution conditions.
