Adipogenesis

Adipogenesis is the biological process through which precursor cells differentiate into mature adipocytes, the cells specialized for lipid storage and endocrine signaling. In genetics, this process is controlled by coordinated changes in gene expression, including activation of transcription factors such as PPARγ and C/EBP proteins, which establish adipocyte identity and promote lipid accumulation. Studying adipogenesis helps clarify how genetic and cellular programs regulate adipose tissue development, energy balance, and metabolic function. These insights support research into obesity, insulin resistance, diabetes, and potential strategies for tissue engineering or therapeutic modulation of fat-cell formation.

Expansion and Adipogenesis Induction of Adipocyte Progenitors from Perivascular Adipose Tissue Isolated by Magnetic Activated Cell Sorting

Here we report a method for isolation of Adipocyte Progenitor Cell (APC) populations from Perivascular Adipose Tissue (PVAT) using Magnetic-activated Cell Sorting (MCS). This method allows for an increased isolation of APC per gram of adipose tissue when compared to Fluorescence-Activated Cell Sorting (FACS).

Exogenous mitochondria of human and murine origin can be internalized by differentiating preadipocytes and tracked throughout adipogenic differentiation using imaging and genetic tools. While these mitochondria physically interact with the endogenous mitochondrial network, they do not increase the expression of markers associated with mature brown adipocytes.

This video demonstrates the differentiation of bone marrow stromal cells (BMSCs) into adipocytes. In the two-step process, in the presence of specific growth factors and hormones, undifferentiated BMSCs first differentiate into preadipocytes. The second step involving treatment with glucose and insulin leads to the formation of mature adipocytes with multiple lipid droplets.

An Enzymatic Method to Rescue Mesenchymal Stem Cells from Clotted Bone Marrow Samples

Mesenchymal stem cells are usually obtained from bone marrow and require expansion culture. When samples clot before processing, a protocol using the (enzymatic) thrombolytic drug urokinase can be applied to degrade the clot. Thus, cells are released and available for expansion culture. This protocol provides a rapid and inexpensive alternative to resampling.

Isolation and Differentiation of Stromal Vascular Cells to Beige/Brite Cells

Primary white preadipocytes isolated from white adipose tissues in mice can be differentiated into beige/brite cells. Presented here is a reliable cellular model system to study the molecular regulation of "browning" of white fat.
