Antigen Labeling

Antigen labeling is a technique that attaches a detectable tag to an antigen or to an antibody that specifically binds it, enabling the target to be visualized, measured, or tracked. Labels such as fluorescent dyes, enzymes, biotin, or nanoparticles generate signals when the labeled molecule binds its complementary partner through specific antigen–antibody interactions. In immunology and infection research, antigen labeling supports immunofluorescence microscopy, flow cytometry, immunoassays, and pathogen localization. These approaches help researchers identify immune targets, characterize cellular responses, quantify antigen expression, and evaluate diagnostic or therapeutic strategies with greater sensitivity and spatial resolution.

Antigen Specific In Vivo Killing Assay using CFSE Labeled Target Cells

Many infections elicit a strong CTL response, but occasionally, the quantity of responding cells does not correlate to control of the pathogen1. One measure of CTL quality is their ability to kill specifically2. CFSE labeling of target cells can be used to investigate this CTL response quality in vivo3,4.

Development of an Antigen-driven Colitis Model to Study Presentation of Antigens by Antigen Presenting Cells to T Cells

In this antigen-driven colitis model, OT-II CD4+ T cells expressing a red fluorescent protein were adoptively transferred into RAG-/- mice that express a green fluorescent protein in mononuclear phagocytes (MPs). The hosts were challenged with Escherichia coli (E.coli) expressing the ovalbumin protein (OVA) fused to a cyan fluorescent protein (CFP).

Metabolic Labeling

2023

Metabolic labeling is used to probe the biochemical transformations and modifications that occur in a cell. This is accomplished by using chemical analogs that mimic the structure of natural biomolecules. Cells utilize analogs in their endogenous biochemical processes, producing compounds that are labeled. The label allows for the incorporation of detection and affinity tags, which can then be used to elucidate metabolic pathways using other biochemical analytical techniques, such as SDS-PAGE...
