Peripheral Blood Lymphocytes

Peripheral blood lymphocytes are immune cells circulating in the bloodstream, primarily T cells, B cells, and natural killer cells, that coordinate adaptive and innate defenses against infection. T and B cells recognize specific antigens through antigen receptors, while natural killer cells detect and destroy stressed or infected cells; activation triggers cytokine release, clonal expansion, antibody production, or cytotoxic responses. In immunology and infection research, these cells can be isolated from blood using density-gradient separation and analyzed with flow cytometry, culture, or functional assays. Their abundance, phenotype, and activity help characterize immune status, pathogen-specific responses, vaccine effects, and disease progression.

This article describes a method for the generation and propagation of human T cell clones that specifically respond to a defined alloantigen. This protocol can be adapted for cloning human T cells specific for a variety of peptide-MHC ligands.

Expansion of Human Peripheral Blood γδ T Cells using Zoledronate

2011

A method to expand γδ T cells from peripheral blood mononuclear cells (PBMC) is described. PBMC-derived γδ T cells are stimulated and expanded using zoledronate and interleukin-2 (IL-2). Large scale expansion of γδ T cells can be applied to autologous cellular immunotherapy of cancer.

Ex Vivo Expansion of Natural Killer Cells from Peripheral Blood Mononuclear Cells

2025

This video demonstrates a technique for the expansion of natural killer (NK) cells directly from peripheral blood mononuclear cells (PBMCs) using a feeder cell line. PBMCs are co-cultured with genetically modified lymphoblastoid feeder cells expressing membrane-bound interleukin-21 (mIL-21), and the culture is supplemented with the cytokines interleukin-2 (IL-2) and interleukin-15 (IL-15). Together, this cocktail of ligands leads to the prolonged expansion of NK cells with high purity.
