Serotype Determination

Serotype determination is the identification of distinct antigenic variants within a species of pathogen, an important process in immunology and infection research because these variants can differ in transmission, immune recognition, and clinical relevance. The method typically exposes a pathogen or clinical sample to serotype-specific antibodies or molecular markers and assigns the serotype according to characteristic antigen-antibody reactions or genetic signatures. In infectious disease surveillance, serotype determination supports outbreak investigation, epidemiological tracking, vaccine development, and evaluation of vaccine coverage. It also helps researchers compare circulating strains and assess how changes in surface antigens may influence host immunity and diagnostic accuracy.

Latex agglutination testing is a simple, rapid and inexpensive method for serotyping Streptococcus pneumoniae, and has also been widely applied in diagnostic microbiology. This manuscript describes the in-house production of latex agglutination reagents, quality control procedures and the application of this technique to pneumococcal serotyping.

Here, we present a protocol to generate a proof-of-principle divalent adenovirus type 5 (Ad5) vector Ad5/H5-HVR1-KWAS-HVR5-His6 by utilizing the Antigen Capsid-Incorporation strategy. This vector was demonstrated to exhibit qualitative fitness, the capability to escape Ad5-positive sera in vitro, and the antigenicity as well as immunogenicity to the incorporated antigens.

Systemic Delivery of MicroRNA Using Recombinant Adeno-associated Virus Serotype 9 to Treat Neuromuscular Diseases in Rodents

Here we describe the delivery of microRNA using a recombinant adeno-associated virus serotype 9 in a mouse model of a neuromuscular disease. A single peripheral administration in mice resulted in sustained miRNA overexpression in muscle and motor neurons, providing an opportunity to study miRNA function and therapeutic potential in vivo.
