Viable Cells

Viable cells are living cells that retain intact membranes, metabolic activity, and the capacity to perform biological functions, making them essential for studying immune responses and infectious disease. Cell viability is commonly assessed by measuring membrane integrity or metabolic activity, such as whether cells exclude vital dyes or convert indicator substrates through active cellular enzymes. In immunology and infection research, viable immune or host cells help reveal pathogen entry, replication, cytotoxicity, inflammation, and antimicrobial responses under controlled experimental conditions. Accurate viability measurements improve interpretation of cell-based assays, support culture quality, and distinguish direct biological effects from damage caused by handling or experimental treatments.

A protocol to utilize a poly(N-iso-propylacrylamide) (PIPAAm) coated microfilter for effective capture and thermoresponsive release of viable circulating tumor cells (CTC) is presented. This method allows capture of CTC from patients' blood and subsequent release of viable CTC for downstream off-chip culture, analyses and characterization.

Circulating tumor cells are isolated from the blood of cancer patients without inflicting cellular damage. Isolation of tumor cells is accomplished using a bimolecular surface of E-selectin in addition to antibodies against epithelial markers. A nanotube coating specifically promotes cancer cell adhesion resulting in high capture purities.

Assessing Root Colonization of Tomato Seedlings by a Pathogenic Bacterium Using Viable Cell Counts

Source: Matthysse, A. G. Adherence of Bacteria to Plant Surfaces Measured in the Laboratory. J. Vis. Exp., (2018)This video demonstrates a protocol for measuring root colonization of tomato seedlings by wild-type and cellulose-deficient pathogenic bacteria. Viable counts of tightly adhering bacteria show greater adhesion and microcolony formation by the wild-type strain, highlighting cellulose’s role in stable root colonization.
