Donor-derived Hematopoietic Tissue

Donor-derived hematopoietic tissue is blood-forming tissue obtained from another individual and used to replace or restore a recipient’s hematopoietic system. It contains hematopoietic stem and progenitor cells, collected from bone marrow, peripheral blood, or umbilical cord blood, that home to the recipient’s marrow and generate new blood and immune cells after transplantation. Clinical success depends on factors such as HLA compatibility, conditioning treatment, cell dose, and donor-recipient immune interactions. This approach treats disorders including leukemia, lymphoma, aplastic anemia, and inherited blood diseases, while also requiring monitoring for graft-versus-host disease, infection, graft failure, and other complications.

Quantification of donor-derived cells is required to monitor engraftment after stem cell transplantation in patients with hemoglobinopathies. A combination of flow cytometry-based cell sorting, colony formation assay, and subsequent analysis of short tandem repeats may be used to assess the proliferation and differentiation of progenitors in the erythroid compartment.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

Mechanical microkeratome dissection to prepare tissue results in thinner, more symmetric grafts with less endothelial cell loss and improved outcomes.
