Endothelial Cell Harvest

Endothelial cell harvest is the process of isolating endothelial cells from blood vessels or vascular tissues for laboratory study and medical research. During harvest, tissue is mechanically dissected and enzymatically digested to loosen cell–cell and cell–matrix contacts, after which centrifugation, selective adhesion, or culture conditions help enrich the endothelial population. Harvested cells support studies of vascular biology, angiogenesis, inflammation, and endothelial responses to drugs or biomaterials. Reliable isolation and expansion provide physiologically relevant models for investigating vascular disease, evaluating therapeutic strategies, and developing tissue-engineered blood vessels.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.

This protocol describes the removal and purification of pulmonary artery endothelial cells (PAECs) that adhere to the balloon tips of Swan-Ganz catheters while they are wedged in the pulmonary artery during right heart catheterization and remain adherent to the deflated balloon following removal from the patient's body.

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.
