Neun Negative Sorting

NeuN-negative sorting is a cell-separation method that enriches brain cells lacking NeuN, a nuclear marker commonly associated with mature neurons. In this approach, tissue is dissociated, NeuN-positive nuclei or cells are identified with a NeuN-specific antibody, and fluorescence-activated or related sorting methods collect the unlabeled fraction as the NeuN-negative population. This fraction can contain non-neuronal cells, including glial and other supporting cell types, depending on the tissue and experimental design. In neuroscience, NeuN-negative sorting helps separate cellular populations for gene-expression, epigenetic, and functional analyses, improving the resolution of studies of brain organization and disease.

Fluorescence Activated Cell Sorting of Plant Protoplasts

A method for isolating specific cell types from plant material is demonstrated. This technique employs transgenic marker lines expressing fluorescent proteins in particular cell types, cellular dissociation and Fluorescence Activated Cell Sorting. Additionally, a growth setup is established here that facilitates treatment of Arabidopsis thaliana seedlings prior to cell sorting.

Fluorescence-Activated Nuclei Negative Sorting of Neurons Combined with Single Nuclei RNA Sequencing to Study the Hippocampal Neurogenic Niche

Presented here is a method to sequence single nuclei isolated from the mouse dentate gyrus that excludes most neurons through fluorescence-activated nuclei (FAN)-sorting. This approach generates high-quality expression profiles and facilitates the study of most other cell types represented in the niche, including scarce populations such as neural stem cells.
