Mice have been used as a model for studying many forms of transplantation, including corneal transplantation. We describe in this report a murine model for both acute and late-term corneal transplantation.
Corneal transplantation is the most common form of organ transplantation in the United States with between 45,000 and 55,000 procedures performed each year. While several animal models exist for this procedure and mice are the species that is most commonly used. The reasons for using mice are the relative cost of using this species, the existence of many genetically defined strains that allow for the study of immune responses, and the existence of an extensive array of reagents that can be used to further define responses in this species. This model has been used to define factors in the cornea that are responsible for the relative immune privilege status of this tissue that enables corneal allografts to survive acute rejection in the absence of immunosuppressive therapy. It has also been used to define those factors that are most important in rejection of such allografts. Consequently, much of what we know concerning mechanisms of both corneal allograft acceptance and rejection are due to studies using a murine model of corneal transplantation. In addition to describing a model for acute corneal allograft rejection, we also present for the first time a model of late-term corneal allograft rejection.
Korneal transplantasjon er en av de mest vellykkede og vanlige typer transplantasjon utført i mennesker. Årsakene til at denne operasjonen er utført er et resultat av skade, smittsomme sykdommer 1, eller andre former for ikke-smittsomme hornhinnesykdommer 2. Tall fra Eye Bank Association of America viser at over 46.000 ble utført i 2011 (se nettside på: restoresight.org/eye_banks/eye_banks.html). Faktorer ansvarlig for hornhinner status som et immun privilegium nettstedet inkluderer mangel på både blod og lymfeårer i hornhinnen, en slektning fravær av antigenpresenterende celler, faktorer produsert av hornhinnen som suppress immuneffektorceller funtions 9-15, lav ekspresjon av MHC-antigener 16, og ekspresjonen av Fasl 17-20.
Men på tross av disse faktorer som disponerer disse transplantater for suksess, har de gjennomgår avvisning 3-7. Følgelig forstå de mekanismer som medierer denne avvisningen, samt å teste ulike behandlinger for å forhindre avstøtning er av avgjørende betydning. For dette formål, beskriver vi her en murin modell av korneal transplantasjon som har vært i bruk i over 20 år for å studere korneal transplantasjon i et kontrollert miljø eksperimentelt. Siden transplantasjons svar involvere mange ulike faktorer som arbeider i konsert som vil ultimate avgjør om det transplanterte vevet mislykkes eller lykkes, er det ikke mulig å forstå betydningen av disse faktorene i noen in vitro modell. Følgelig er studier med intakte dyr nødvendig for å fastslå hvilke faktorer som er viktige for enten suksess eller failure av transplantert vev.
Mens andre arter av dyr har blitt brukt til å studere korneal transplantasjon, har den murine modellen flere fordeler sammenlignet med bruk av andre arter. Den første er at det finnes mange stammer av mus som uttrykker visse transgener eller har vært gen-målrettede å mangle uttrykk for spesifikke immunologiske faktorer som har funksjon i transplantasjon kan bedre undersøkt. I tillegg er det mange reagenser (både rekombinante faktorer og antistoffer som nøytraliserer faktorer) som er spesifikke for mus og som ikke finnes for mange andre arter av dyr. På grunn av eksistensen av disse faktorene, har denne modellen vært brukt mye til å identifisere relevante faktorer involvert i akutte hornhinnen leverallograft svar 15, 17,18,20 -29. Videre er mange av de faktorene som er involvert i hornhinnetransplantasjon også kjent for å være funksjonelle i transplantasjon av andre vev.
Muse-modell av hornhinnetransplantasjon beskrevet her gjør at etterforsker for å studere menneskelig hornhinnen rejeksjon ved en modell som er prediktive for hvilke faktorer som er best assosiert med både avvisning 15,17,18,20, 26-30 og aksept 21-25 av hornhinnen allografter.
