Hydrogel Substrate Preparation

Hydrogel substrate preparation is the process of producing water-rich, polymer-based materials that provide controlled physical and biochemical environments for bioengineering studies. It typically involves dissolving or mixing hydrogel precursors, shaping the material, and inducing crosslinking through chemical, thermal, ionic, or light-based mechanisms; adjusting polymer concentration and crosslink density tunes properties such as stiffness, porosity, swelling, and degradation. Prepared substrates can support cell adhesion, migration, proliferation, and differentiation, making them useful for tissue engineering, regenerative medicine, biomaterials testing, and in vitro models that examine how material properties influence cellular behavior.

We present a new polyacrylamide hydrogel, called hydroxy-PAAm, that allows a direct binding of ECM proteins with minimal cost or expertise. The combination of hydroxy-PAAm hydrogels with microcontact printing facilitates independent control of many cues of the natural cell microenvironment for studying cellular mechanostransduction.

In this video, we demonstrate a high throughput assay for carbohydrate-active enzyme screening using insoluble chromogenic polymer hydrogel substrates. The enzyme degrades these substrates into a soluble colored product that can be quantified by measuring the color intensity of the product.
