Fluorescent Protein Stain

Fluorescent protein stain is a labeling method that makes specific proteins visible by attaching or applying fluorescent molecules, enabling sensitive detection in biological samples. When illuminated at an appropriate excitation wavelength, the fluorophore absorbs energy and emits light at a longer wavelength, producing a detectable signal that can be imaged or quantified. In biological techniques, fluorescent protein stains support protein localization, expression analysis, and visualization of cellular structures in fixed cells, tissues, gels, or other specimens. Their specificity and optical contrast help researchers investigate molecular interactions, cellular organization, and changes associated with development or disease.

Here, we describe a detailed protocol outlining a new fluorescent staining technique for total protein detection in polyacrylamide gels. The protocol utilizes a silver ion-specific fluorescence turn-on probe, which detects Ag+-protein complexes, and eliminates certain limitations of traditional chromogenic silver stains.
