Enzymatic Steps

Enzymatic steps are the individual enzyme-catalyzed reactions that convert substrates into products within a biochemical pathway, making complex biological processes manageable and regulated. Each step occurs when a substrate binds an enzyme’s active site, where specific chemical groups and, in some cases, cofactors help lower activation energy and drive a defined transformation. The product of one reaction may become the substrate for the next, linking sequential steps in metabolism, DNA replication, and cellular signaling. Mapping these steps helps researchers explain pathway regulation, identify effects of genetic or chemical disruption, and design experiments or therapies that target particular reactions.

Rapid One-step Enzymatic Synthesis and All-aqueous Purification of Trehalose Analogues

Trehalose analogues are emerging as important molecules for bio(techno)logical and biomedical applications. We describe an optimized protocol for enzymatically synthesizing and purifying trehalose analogues that is simple, efficient, fast, and environmentally friendly. Its application to the rapid production and administration of a probe for the detection of mycobacteria is demonstrated.

Class 1 histone deacetylases (HDACs) like RpdA have gained importance as potential targets to treat fungal infections. Here we present a protocol for the specific enrichment of TAP-tagged RpdA combined with an HDAC activity assay that allows in vitro efficacy testing of histone deacetylase inhibitors.

Here we describe a method for the isolation of hepatic stellate cells from mouse liver. For stellate cell purification, mouse livers are digested in situ and in vitro by pronase-collagenase treatment prior to density gradient centrifugation. This technique yields highly pure hepatic stellate cells.

A Step by Step Protocol for Subretinal Surgery in Rabbits

Retinal pigment epithelium (RPE) replacement strategies and gene-based therapy are considered for several retinal degenerative conditions. For clinical translation, large eye animal models are required to study surgical techniques applicable in patients. Here we present a rabbit model for subretinal surgery geared towards RPE transplantation, which is versatile and cost-efficient.
