Immunosuppressive Agents

Immunosuppressive agents are drugs or biological compounds that reduce immune-system activity, helping control harmful inflammation and immune responses against transplanted tissues or the body’s own cells. They act through mechanisms such as blocking immune-cell signaling, limiting lymphocyte activation and proliferation, or reducing the production and action of inflammatory mediators. In biology and medicine, these agents support organ and tissue transplantation, treatment of autoimmune and inflammatory disorders, and research into immune regulation. Their effects can improve disease control or graft survival, but excessive suppression may increase susceptibility to infection and complicate long-term treatment decisions.

Here we describe a high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) assay to quantify the immunosuppressant tacrolimus in dried blood spots using a simple manual protein precipitation step and online column extraction.

This protocol describes the preparation and characterization of a dendrimeric magnetic resonance imaging (MRI) contrast agent that carries cyclen-based macrocyclic chelates coordinating paramagnetic gadolinium ions. In a series of MRI experiments in vitro, this agent produced an amplified MRI signal when compared to the commercially available monomeric analogue.

Using optimized human peripheral blood mononuclear cell (PBMC) ex vivo assays, we showed that a combustible tobacco product preparation markedly suppresses receptor-mediated intracellularly secreted cytokines and cytolytic ability of effector PBMCs. These rapid assays may be useful in product evaluation and understanding the potential long-term effects of tobacco exposure.
