Primary Productivity

Primary productivity is the rate at which autotrophic organisms convert inorganic carbon into organic matter, providing the energy that supports most ecosystems. In aquatic and terrestrial environments, plants, algae, and some bacteria use photosynthesis to fix carbon dioxide into biomass, whereas chemosynthetic microbes obtain energy by oxidizing inorganic compounds in the absence of light. Scientists distinguish gross primary productivity, the total carbon fixed, from net primary productivity, the biomass remaining after autotrophic respiration. These measures help assess ecosystem function, compare habitats, and trace carbon movement through food webs and the global carbon cycle.

The total amount of energy acquired by primary producers in an ecosystem is called gross primary production (GPP). However, of this energy, producers use some for metabolic processes, and some is lost as heat, decreasing the amount of energy available to the next trophic level. The remaining usable amount of energy is called the net primary productivity (NPP). In terrestrial ecosystems, NPP is driven by climate, while light penetration and nutrient availability drive NPP in aquatic ecosystems.

Primary human bronchial epithelial cells are difficult to transduce. This protocol describes the production of lentiviruses and their concentration as well as the optimal culture conditions necessary to achieve highly efficient transductions in these cells throughout differentiation to a pseudostratified epithelium.

The primary cilium is fundamentally important in neural progenitor cell proliferation, neuronal differentiation, and adult neuronal function. Here, we describe a method to study ciliogenesis and the trafficking of signaling proteins to cilia in neural stem/progenitor cells and differentiated neurons using primary neurosphere cultures.
