Rat Neurodegenerative Models

Rat neurodegenerative models are laboratory systems that reproduce selected features of human disorders involving progressive neuronal dysfunction and loss, providing a bridge between cellular studies and clinical research. Researchers create these models through genetic modifications, administration of neurotoxic compounds, targeted brain injury, or other manipulations that disrupt defined neural pathways and produce measurable behavioral, molecular, or anatomical changes. These models support investigation of disease mechanisms, including protein accumulation, neuroinflammation, synaptic dysfunction, and neuronal death, while enabling evaluation of biomarkers and potential therapies. Their physiological and behavioral complexity also helps assess treatment effects before clinical testing, although differences between rats and humans require careful interpretation.

We present an optimized inexpensive and reliable negative geotaxis assay in Drosophila melanogaster as a model for neurodegenerative disorders. Being more sensitive to mild locomotor defects, this assay will help screen for potential genetic interactions and drug targets.

We describe a methodology combining automated cell culturing with high-content imaging to visualize and quantify multiple cellular processes and structures, in a high-throughput manner. Such methods can aid in the further functional annotation of genomes as well as identify disease gene networks and potential drug targets.
