Tail Diameter Measurement

Tail diameter measurement is the quantitative assessment of the width of a biological tail or tail-like structure, providing an objective indicator of morphology, development, and structural variation. In biology, researchers typically obtain calibrated images or microscopy measurements, define consistent anatomical reference points, and determine diameter using a scale bar, imaging software, or cross-sectional analysis while controlling for orientation and magnification. This measurement supports comparisons among organisms, cells, tissues, or experimental groups and can reveal changes associated with growth, injury, disease, or genetic and environmental conditions. Standardized measurements improve morphological analysis and help connect structural differences with biological function.

The tail-suspension test is validated as an experimental procedure to assess antidepressant efficacy of drug treatments in mice. Mice are suspended by their tails for six minutes and escape-related behaviors are assessed. We describe procedures used in conducting the tail suspension test.

Size Matters: Measurement of Capsule Diameter in Cryptococcus neoformans

The polysaccharide capsule is the primary virulence factor in Cryptococcus neoformans, and its size correlates with strain virulence. Capsule diameter measurements are used in phenotypic testing and to gauge therapeutic efficacy. Here a standard method of capsule induction is presented, and two methods of staining and measuring diameter are compared.
