Trna Charging Levels

tRNA charging levels are the proportions of cellular transfer RNAs (tRNAs) carrying their corresponding amino acids, a measure that links nutrient status to protein synthesis. Aminoacyl-tRNA synthetases use ATP to attach a specific amino acid to the tRNA’s 3′ end; changes in amino acid availability, enzyme activity, or tRNA demand shift the balance between charged and uncharged forms. Measuring these levels helps researchers assess translational capacity and cellular responses to nutrient limitation, stress, and growth conditions, while comparisons among treatments can reveal how cells regulate gene expression through translation and maintain accurate protein production.

Here we present a method for directly measuring transfer RNA charging levels from purified Escherichia coli RNA as well as a way to compare relative levels of transfer RNA, or any other short RNA, across different samples based on the addition of spike-in cells expressing a reference gene.

This protocol describes the preparation of a synthetic tRNA substrate for the Entamoeba histolytica DNA/tRNA methyltransferase 2 (Dnmt2) homolog Ehmeth and the measure of its methyltransferase activity. This experimental approach can be used for investigating the activity of other Dnmt2 proteins.
