Colon Epithelial Cells

Colon epithelial cells are the specialized cells lining the inner surface of the colon, where they regulate barrier integrity, nutrient and water absorption, and interactions with the gut microbiome. Organized into crypts, these cells include absorptive colonocytes, mucus-producing goblet cells, and other lineages that arise from intestinal stem cells and maintain the epithelium through continuous renewal; disruption of signaling pathways controlling proliferation, differentiation, or cell death can promote tumor formation. In cancer research, colon epithelial cells support studies of colorectal cancer initiation, tumor progression, genetic alterations, drug responses, and interactions with inflammatory or microbial factors, improving disease models and therapeutic development.

We present a protocol that combines cell isolation and whole-cell patch-clamp recording to measure the electrical properties of the primary dissociated epithelial cells from the rat cauda epididymides. This protocol allows for investigation of the functional properties of primary epididymal epithelial cells to further elucidate the physiological role of the epididymis.

Electrophysiological Recording from Colonic Afferent Nerves in an Ex Vivo Rat Colon

This video demonstrates a method to record the activity of colonic afferent nerves in an ex-vivo rat colon, cannulated at both ends and infused with saline to increase intraluminal pressure. As pressure increases, the nerve signal rises, indicating its stimulation.
