Primary Tumor Culture

Primary tumor culture is the laboratory maintenance and study of cells freshly isolated from a patient’s tumor, preserving features that may be lost in long-established cell lines. Researchers typically dissociate tumor tissue into viable cells, provide defined growth conditions, and support attachment or three-dimensional organization while monitoring cell survival and proliferation. In cancer research, these cultures can retain aspects of tumor heterogeneity and interactions among malignant, stromal, and immune cell populations, making them useful for investigating tumor biology, treatment response, and drug resistance. They also support patient-derived models that can complement animal studies and inform more individualized approaches to cancer therapy.

In vitro Organoid Culture of Primary Mouse Colon Tumors

A simple method to establish primary murine colon tumor organoid is described. This method utilizes the feature that colon tumor cells survive and grow into organoids in media containing limited growth factors, whereas normal colon epithelial do not.

The primary cilium is fundamentally important in neural progenitor cell proliferation, neuronal differentiation, and adult neuronal function. Here, we describe a method to study ciliogenesis and the trafficking of signaling proteins to cilia in neural stem/progenitor cells and differentiated neurons using primary neurosphere cultures.

Vestibular Schwannomas (VSs) are non-malignant tumors of Schwann cell (SC) origin, associated with mutations in the NF2 tumor suppressor gene. We report a reproducible, efficient protocol for primary human VS cell culture that allows for molecular and cellular experimental manipulation and analysis and recapitulates the heterogeneous nature of human disease.

A protocol is described to extract primary human cells from surgical colon tumor and normal tissues. The isolated cells are then cultured on soft elastic substrates (polyacrylamide hydrogels) functionalized by an extracellular matrix protein, and embedded with fluorescent microbeads. Traction cytometry is performed to assess cellular contractile stresses.
