Skin Reconstruction Model

A skin reconstruction model is a laboratory system that recreates key structural and biological features of human skin to study tissue function, injury, and disease, including cancer. These models typically combine epidermal keratinocytes with dermal fibroblasts and an extracellular-matrix scaffold, often at an air–liquid interface that supports keratinocyte differentiation and formation of a stratified epidermis. In cancer research, reconstructed skin can provide a controlled setting for examining tumor initiation, invasion, interactions between cancer cells and surrounding stroma, and responses to candidate treatments. By complementing animal models and conventional two-dimensional cultures, it can support mechanistic studies and more physiologically relevant drug evaluation.

In this report, we describe the three-dimensional skin reconstruct model which mimics human skin in architecture and composition. Melanocyte physiology, melanoma progression and the fate of dermal stem cells have been investigated using the skin reconstruct model. The model is also useful as a preclinical tool for drug assessment.

A Murine Model of a Burn Wound Reconstructed with an Allogeneic Skin Graft

The aim of this study was to develop a murine model of burn wound healing. A thermal burn was induced on the dorsal skin of mice using a preheated brass template. Burned tissue was debrided and overlaid with a skin graft harvested from the tail of a genetically similar donor mouse.

Chemical-induced Two-stage Skin Carcinogenesis Model: An Experimental In Vivo Mouse Model of Skin Cancer

This video describes the procedure to induce two-stage skin carcinogenesis to study tumor initiation by topical application of chemicals. In the first step, a mutagen, DMBA, is applied onto the skin of a mouse model which initiates tumor formation by causing mutations in the stem cells, followed by the application of a growth stimulator, TPA, which accelerates skin papilloma formation.
