Glycobiology

Glycobiology is the study of carbohydrates, glycoconjugates, and the molecular information encoded by their structures, making it central to understanding biological recognition and regulation. In cells, sugars attach to proteins, lipids, or other carbohydrates through glycosidic bonds, while glycosyltransferases build these structures and glycosidases selectively remove or remodel them; differences in linkage, branching, and composition can alter molecular interactions. In chemistry and related fields, glycobiology supports the analysis and synthesis of complex glycans, investigation of cell signaling and host-pathogen interactions, and development of biomarkers, vaccines, and carbohydrate-based therapeutics. Its methods also advance research into how altered glycosylation contributes to disease.

Models of Murine Vaginal Colonization by Anaerobically Grown Bacteria

The protocol presents a mouse model of vaginal colonization with anaerobically cultured human vaginal bacteria. We focus on Gardnerella vaginalis, while including suggestions for Prevotella bivia and Fusobacterium nucleatum. This protocol can also be used as a guide for vaginal inoculations and viable recovery of other anaerobically grown bacteria.

A modular approach to the synthesis of N-glycans for attachment to an aluminum oxide-coated glass slide (ACG slide) as a glycan microarray has been developed and its use for the profiling of an HIV broadly neutralizing antibody has been demonstrated.

Sialic acid is a typical monosaccharide-unit found in glycoconjugates. It is involved in a plethora of molecular and cellular interactions. Here we present a method to modify cell surface sialic acid expression using metabolic glycoengineering with N-acetylmannosamine derivatives.
