Low Oxygen Incubation

Low Oxygen Incubation is a laboratory technique that maintains cells, tissues, or embryos in an atmosphere with less oxygen than standard culture conditions, helping researchers model physiological or developmental environments. A hypoxia-controlled incubator regulates oxygen concentration, often alongside carbon dioxide, temperature, and humidity, to limit oxygen availability and produce reproducible culture conditions. In developmental biology, this approach allows investigators to examine how oxygen levels influence cell differentiation, proliferation, tissue patterning, and embryo development. Comparing low-oxygen and atmospheric-oxygen cultures can reveal oxygen-sensitive signaling pathways and clarify how changing oxygen environments contribute to normal development or developmental abnormalities.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
