Fetal Fibroblasts

Fetal fibroblasts are connective-tissue cells from developing embryos or fetuses that produce extracellular matrix and support tissue formation, making them valuable for studying development and inherited disease. They proliferate in culture and synthesize structural proteins such as collagen, while developmental signals regulate their gene expression, growth, migration, and matrix organization. In genetics, cultured fetal fibroblasts provide a living cellular system for analyzing gene expression, chromosome abnormalities, mutations, and responses to experimental perturbations. Their developmental origin also supports research on tissue differentiation, genetic disorders, and therapeutic strategies, although findings may differ from those in adult cells.

Fetal and perinatal death is a common feature when studying genetic alterations affecting cardiac development. High-frequency ultrasound imaging has improved 2-D resolution and can provide excellent information on early cardiac development and is an ideal method to detect the impact on cardiac structure and function prior to death.

This protocol provides a comprehensive procedure to isolate, expand, and immortalize fibroblasts from radical prostatectomies. Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

We describe examination of fetal cardiac function with contemporary functional fetal echocardiography and fetoplacental Doppler ultrasound using the VisualSonics VEVO 2100 microultrasound in a surgically induced model of intrauterine fetal growth restriction in a rabbit.
