Ribonucleoprotein Electroporation

Ribonucleoprotein electroporation is a genome-editing technique that introduces preassembled protein–RNA complexes into cells using brief electrical pulses, enabling targeted genetic modification without delivering DNA. The electrical field temporarily permeabilizes the cell membrane, allowing Cas9 ribonucleoproteins, composed of the Cas9 nuclease and guide RNA, to enter the cytoplasm and nucleus, where the guide directs sequence-specific DNA cleavage. Cellular repair of these breaks can generate targeted insertions or deletions, while a supplied donor template can support precise sequence replacement. In genetics research, the method enables rapid gene knockout, functional screening, and engineered cell production with transient reagent exposure and reduced risk of vector integration.

This protocol describes techniques for isolating primary mouse hepatocytes from the liver and electroporating CRISPR-Cas9 as ribonucleoproteins and mRNA to disrupt a therapeutic target gene associated with an inherited metabolic disease of the liver. The methods described result in high viability and high levels of gene modification after electroporation.
