Actin Protrusion

Actin protrusion is the formation of dynamic, actin-rich extensions that reshape a cell’s surface and enable directed movement, making it central to immune surveillance and infection biology. At leading edges, actin filaments polymerize against the plasma membrane; branching and cross-linking, regulated by nucleation-promoting factors and actin-binding proteins, generate protrusive force, while membrane adhesion stabilizes the advancing structure. In leukocytes, these protrusions support chemotaxis, antigen sampling, cell-cell interactions, and phagocytosis, whereas pathogens can manipulate actin remodeling to enter or spread between cells. Visualizing or perturbing this process connects cytoskeletal dynamics with host defense, inflammatory responses, and infection mechanisms, informing studies of immune dysfunction and potential therapeutic targets.

Study of the Actin Cytoskeleton in Live Endothelial Cells Expressing GFP-Actin

Microscopic imaging of live endothelial cells expressing GFP-actin allows characterization of dynamic changes in cytoskeletal structures. Unlike techniques that use fixed specimens, this method provides a detailed assessment of temporal changes in the actin cytoskeleton in the same cells before, during, and after various physical, pharmacological, or inflammatory stimuli.

Filamentous actin (F-actin) plays an important role in spinogenesis, synaptic plasticity, and synaptic stability. Quantification of F-actin puncta is therefore a useful tool to study the integrity of synaptic structures. This protocol describes the procedures of quantifying F-actin puncta labeled with Phalloidin in low-density primary cortical neuronal cultures.
