Suit Doffing

Suit doffing is the controlled removal of protective clothing and equipment after work in a potentially contaminated environment, helping prevent exposure and the spread of infectious agents. The procedure uses a deliberate sequence that limits contact with contaminated outer surfaces, incorporates hand hygiene or disinfection between steps, and separates used items from clean areas. In immunology and infection research, correct doffing protects laboratory personnel, patients, and surrounding environments from cross-contamination. Standardized training and careful technique support biosafety, reduce occupational risk, and help maintain the integrity of infectious disease investigations and other procedures involving biological hazards.

The sequence of removing or doffing PPE starts with the gloves, as they are the most contaminated. Next is removal of the face shield or goggles, as they would interfere with removing other PPE. Then remove the gown, followed by the mask or respirator. Perform hand hygiene between steps if hands become contaminated and immediately after removing all PPE. Generally, the outside front and sleeves of the isolation gown, the goggles or the mask, the respirator, and the face shield are contaminated.

Biofilm infections show high tolerance towards chemotherapy. No single assay captures the complexity of biofilms. Instead, complementary assays are needed. We present a screening platform (developed for S. aureus) that combines assays for viability, biomass, and biofilm matrix. It allows anti-biofilm drug discovery, including the assessment of long-term chemotherapeutic effects.

Galaxy and DAVID have emerged as popular tools that allow investigators without bioinformatics training to analyze and interpret RNA-Seq data. We describe a protocol for C. elegans researchers to perform RNA-Seq experiments, access and process the dataset using Galaxy and obtain meaningful biological information from the gene lists using DAVID.
