Automated Quantitative Analysis

Automated quantitative analysis is a computational method that converts complex scientific or clinical data into reproducible numerical measurements, reducing reliance on manual assessment. In medicine, software processes standardized images, laboratory results, or other datasets by detecting relevant features, extracting variables, applying predefined rules or statistical models, and performing quality checks. This approach supports medical imaging, digital pathology, biomarker evaluation, disease classification, and monitoring of treatment response. By improving measurement consistency and enabling analysis of large datasets, automated quantitative analysis can strengthen clinical research, reveal subtle patterns, and support more objective decision-making when validated against appropriate reference standards.

Quantitative and Automated High-throughput Genome-wide RNAi Screens in C. elegans

We describe a protocol using C. elegans and RNAi feeding libraries that allows automated measurement of multiple parameters such as fluorescence, size and opacity of individual worms in a population. We give one example of a screen to identify genes involved in anti-fungal innate immunity in C. elegans.

QFA estimates the effect of genetic mutations, drugs or other applied treatments on microbe growth. Salmonella invades and replicates inside intestinal epithelial cells both in Salmonella-specific vacuoles and free in the cytosol (hyper-replication). A high-throughput fluorescence microscopy-based protocol is described here to quantify the intracellular phenotypes of Salmonella by two complementary image analyses through ImageJ, reaching single-cell resolution and scoring.
