Bone Healing Mice

Bone healing mice refers to mouse models used to investigate how bone repairs after injury and how therapies can improve that process. Following a standardized fracture or bone defect, researchers track coordinated inflammation, formation of a cartilage or bone callus, vascularization, and remodeling, while genetic, molecular, imaging, and histological analyses reveal the cells and signals involved. These models help connect mechanisms of skeletal repair with outcomes such as restored bone structure and strength, supporting research on osteoporosis, impaired healing, biomaterials, and regenerative medicine. Their small size, defined genetics, and experimental flexibility make them valuable for preclinical studies, although findings require careful translation to human patients.

The murine closed femoral fracture model is a powerful platform to study fracture healing and novel therapeutic strategies to accelerate bone regeneration. The goal of this surgical protocol is to generate unilateral closed femoral fractures in mice using an intramedullary steel rod to stabilize the femur.

This protocol describes a procedure for serial sampling of femoral bone marrow (BM) without requiring the sacrifice of mice. This procedure facilitates longitudinal studies of the BM composition of mice over time and provides serial access to cells within the BM for ex vivo and transplantation studies.

Micro-computed tomography (µCT) is a non-destructive imaging tool that is instrumental in assessing bone structure in preclinical studies, however there is a lack of consensus on µCT procedures for analyzing the bone healing callus. This study provides a step-by-step µCT protocol that allows the monitoring of fracture healing.
