Fibronectin Extra Domain A

Fibronectin Extra Domain A (EDA) is an alternatively spliced segment of fibronectin, an extracellular matrix glycoprotein that helps organize tissues and regulate cell behavior. EDA inclusion changes fibronectin structure and creates binding sites that influence cell adhesion, migration, and signaling through integrins and innate immune receptors such as Toll-like receptor 4. In healthy tissues, EDA-containing fibronectin contributes to embryonic development and wound repair, whereas persistent expression is associated with inflammation, fibrosis, and tumor progression. Studying EDA supports research into tissue remodeling, disease mechanisms, and potential biomarkers or therapeutic targets in medicine.

This protocol details the isolation of chondrocytes, Fibronectin Adhesion Assay-derived Chondroprogenitors (FAA-CPs), and Migratory Chondroprogenitors (MCPs) from human articular cartilage. It covers enzymatic digestion, fibronectin adhesion, and migration-based assays for isolating and characterizing these cells.

Fibronectins Connect Cells with ECM

2023

Fibronectin is an adhesive glycoprotein present in the extracellular matrix of embryogenic and adult tissue. These molecules primarily aid in regulating cell motility and attachment. A fibronectin molecule is composed of two identical polypeptide chains attached to each other by a pair of disulfide bonds at the C-terminal. Both proteoglycans and collagen are attached to fibronectin proteins, which, in turn, are attached to integrin proteins. These integrin proteins interact with transmembrane...

Conservation of Protein Domains Over Different Proteins

2020

Protein domains are small structurally independent units that are part of a single amino acid chain. Although these domains are often structurally independent, they may rely on synergistic effects to perform their functions as part of a larger protein. Protein domains may be conserved within the same organism, as well as across different organisms. A limited set of protein domains often duplicate and recombine during evolution. These domains can be organized in different combinations to form...

Here, four methods for generating testicular organoids from primary neonatal murine testicular cells are described i.e., extracellular matrix (ECM) and ECM-free 2D and 3D culture environments. These techniques have multiple research applications and are especially useful for studying testicular development and physiology in vitro.
