Knockout Animal Rate

Knockout animal rate is the proportion of genetically engineered animals that carry a planned, functional disruption of a specific gene, making it an important measure of gene-targeting efficiency in biomedical research. Researchers calculate it by comparing confirmed knockout animals with the total number of treated embryos, implanted embryos, or resulting offspring, using genotyping and often sequence analysis to verify the intended alteration. In medicine, this rate helps assess the performance of approaches such as CRISPR-based editing and embryonic stem-cell targeting, supports planning for animal production, and indicates how efficiently a model can be established for studying disease mechanisms, drug responses, and therapeutic targets.

Production of Apolipoprotein C-III Knockout Rabbits using Zinc Finger Nucleases

Recent development in gene targeting tools makes production of knockout (KO) rabbits possible. In the present work, we generated five Apolipoprotein (Apo) C-III KO rabbits using Zinc Finger Nucleases (ZFN). This work demonstrated that ZFN is a highly efficient method to produce KO rabbits.
