Lucifer Yellow Permeability Assay

Lucifer Yellow permeability assay is a fluorescence-based method for measuring how readily a small, hydrophilic tracer crosses a cellular barrier, making it useful for evaluating epithelial or endothelial integrity. In the assay, cells form a monolayer separating two chambers, Lucifer Yellow is added to one side, and fluorescence appearing on the opposite side is quantified; increased passage generally indicates greater paracellular leak through disrupted tight junctions. In medicine and biomedical research, the assay helps characterize intestinal, vascular, and blood-brain barrier models, assess barrier injury or inflammation, and evaluate whether drugs or other treatments alter tissue permeability.

We describe a live whole animal quantitative measurement for permeability of the embryonic zebrafish brain. The technique analyzes the ability to retain cerebrospinal fluid and molecules of different molecular weights within the neural tube lumen and quantifies their movement out of the ventricles. This method is useful for determining differences in epithelial permeability and maturation during development and disease.
