Minipig Msc Isolation

Minipig MSC isolation is a laboratory method for obtaining mesenchymal stromal cells from minipig tissues, providing a clinically relevant large-animal source for biomedical research. The process typically combines tissue collection, mechanical mincing, enzymatic dissociation, filtration or centrifugation, and culture conditions that favor the attachment and expansion of adherent stromal cells. Isolated cells support studies of regenerative medicine, tissue repair, cell-based therapies, and disease modeling, while the minipig’s anatomical and physiological similarities to humans can improve preclinical evaluation. Standardized isolation and characterization help researchers assess cell yield, viability, phenotype, and therapeutic potential.

2016

A protocol establishing mesenchymal stem cells (MSCs) isolated from the synovial fluid and bone marrow of minipigs and non-invasively collected using syringe aspiration is presented. Cellular phenotyping was performed using flow cytometry after cell isolation and in vitro cultivation of bone marrow and synovial fluids derived from MSCs.

MitoCeption: Transferring Isolated Human MSC Mitochondria to Glioblastoma Stem Cells

Here, a protocol (MitoCeption) is presented to transfer mitochondria, isolated from human mesenchymal stem cells (MSC), to glioblastoma stem cells (GSC), with the goal of studying their biological effects on GSC metabolism and functions. A similar protocol can be adapted to transfer mitochondria between other cell types.

Subdural Implantation of a Soft Electrocorticography Array for Cortical Electrophysiology Recording in Minipigs

2025

This video demonstrates the implantation of a soft electrocorticography array and the long-term cortical recordings in a minipig. It outlines the steps involved in surgically implanting the array over the minipig's auditory cortex and recording both baseline and sound stimulus-evoked neuronal action potentials.

Production and Administration of Therapeutic Mesenchymal Stem/Stromal Cell (MSC) Spheroids Primed in 3-D Cultures Under Xeno-free Conditions

The therapeutic potential of mesenchymal stem/stromal cells (MSCs) is well-documented, however the best method of preparing the cells for patients remains controversial. Herein, we communicate protocols to efficiently generate and administer therapeutic spherical aggregates or 'spheroids' of MSCs primed under xeno-free conditions for experimental and clinical applications.

MSC Seeded Scaffold Preparation: A Technique to Prepare Scaffold Seeded with Mesenchymal Stem Cells for Surgical Implantation into Murine Model

2025

This video describes the technique of seeding mesenchymal stem cells (MSCs) onto a biodegradable poly(lactic acid) (PLA) scaffold. The prepared scaffolds can be implanted into the surgical resection cavity of the brain from where they may migrate towards tumor foci. Further, these cells can be engineered to express therapeutic proteins to stimulate apoptosis in cancer cells.
