Nanoparticle Complex Formation

Nanoparticle complex formation is the process of assembling nanoscale materials with drugs, nucleic acids, proteins, or other therapeutic components to create functional complexes for medical use. These structures form through interactions such as electrostatic attraction, hydrophobic association, hydrogen bonding, or chemical coupling, with particle size, surface charge, and formulation conditions influencing stability and cargo release. In medicine, nanoparticle complexes can improve the solubility, protection, transport, and cellular delivery of therapeutics, including agents that are otherwise unstable or poorly absorbed. Their design supports targeted delivery strategies, imaging, diagnostics, and the development of more precise treatments.

An In Vitro Technique to Study Inflammasome Complex Formation in Macrophages

This video demonstrates a technique to study NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome formation in primary macrophages via immunofluorescence. The macrophages are first primed with bacterial lipopolysaccharide (LPS) to upregulate NLRP3 protein expression. Exposing the macrophages to nigericin—a potassium ionophore—leads to the activation of NLRP3 and the formation of inflammasomes, which are visualized via immunofluorescence.
