Pulp Canal Calcification

Pulp canal calcification is the pathological or age-related deposition of mineralized tissue within the dental pulp space, progressively narrowing or obliterating the root canal. It commonly develops after dental trauma, inflammation, caries, or restorative procedures, as odontoblast-like cells produce tertiary dentin that reduces the canal’s diameter. Although the tooth may remain functional and asymptomatic, calcification can complicate pulp vitality assessment and make root canal treatment difficult by obscuring canal entrances and increasing the risk of perforation. Careful radiographic evaluation, often supported by three-dimensional imaging and magnification, helps clinicians diagnose the condition, assess its extent, and plan conservative endodontic management.

We demonstrate the use of various microscopy methods that are useful in observing the calcification of a tubeworm, Hydroides elegans, as well as locating and characterizing the first calcified material. Live microscopy and electron microscopy are used together to provide functional and material information that are important in studying biomineralization.

Establishing a Co-Culture of Dental Pulp Cells and Trigeminal Neurons for Cross-Communication

2025

This video showcases a technique for establishing a co-culture of trigeminal neurons and primary dental pulp cells using a transwell filter. In this physically separated condition, mesenchymal cells obtained from dental pulp tissue secrete paracrine signaling molecules that traverse through the pores, promoting the differentiation and migration of neurons towards the mesenchymal cells establishing a co-culture system.
