Arteriolar Diameter Measurement

Arteriolar diameter measurement is a quantitative technique for determining the width of small arteries, providing a direct indicator of vascular tone and local blood-flow regulation in the brain. Using calibrated high-resolution images or video, researchers identify the vessel boundaries and measure lumen width over time, capturing constriction and dilation produced by vascular smooth muscle activity. In neuroscience, these measurements help assess neurovascular coupling during sensory stimulation, neuronal activation, and changes in blood pressure. They also support studies of cerebrovascular disease, pharmacological responses, and mechanisms that link neural activity to hemodynamic signals used in functional imaging.

The polysaccharide capsule is the primary virulence factor in Cryptococcus neoformans, and its size correlates with strain virulence. Capsule diameter measurements are used in phenotypic testing and to gauge therapeutic efficacy. Here a standard method of capsule induction is presented, and two methods of staining and measuring diameter are compared.

We demonstrate a novel arterial ligation model in murine spinotrapezius muscle, including a step-by-step procedure and description of required instrumentation. We describe the surgery and relevant outcome measurements relating to vascular network remodeling and functional vasodilation using intravital and confocal microscopy.
