Cognitive Optical Brain Imaging

Cognitive optical brain imaging is a family of light-based methods used to visualize brain activity and circulation while animals or people perform cognitive tasks, linking neural dynamics to perception, learning, memory, and decision-making. In experimental settings, fluorescent calcium indicators report activity by changing intensity as intracellular calcium rises during neuronal firing, while optical measurements of hemoglobin absorption track blood-flow and oxygenation changes. Microscopy and wide-field approaches provide complementary spatial and temporal information, from cellular populations to distributed cortical networks. These data help researchers test how brain circuits support cognition, identify abnormalities in neurological disease, and evaluate interventions that alter neural function.

We present a protocol that allows investigation of the neural mechanisms mediating the detrimental impact of emotion on cognition, using functional magnetic resonance imaging. This protocol can be used with both healthy and clinical participants.

Using Fiberless, Wearable fNIRS to Monitor Brain Activity in Real-world Cognitive Tasks

Monitoring brain activity outside the lab without physical constraints presents methodological challenges. A fiberless, wearable functional Near Infrared Spectroscopy (fNIRS) system was used to measure brain activity during an ecological prospective memory task. It was demonstrated that this system could be used to monitor brain activity during non-lab based experiments.

We introduced a novel imaging technique called Optical Multilayer Interference Tomography (OMLIT), which enables unbiased imaging of all cells in brain specimens at the mesoscale and can be seamlessly integrated into the imaging workflow of tape-based serial scanning electron microscopy on the same sample.

This video demonstrates the brain's neural activity recording through an optical window in transgenic mice. The mice express a fluorescent protein that enables the detection of neural activities. A small portion of the skull is carefully removed to install an optical window, and the skull is then covered with black cement to block light before imaging.
