Corneal Innervation

Corneal innervation is the dense network of sensory and autonomic nerve fibers that supplies the cornea, supporting vision, ocular protection, and surface homeostasis. Sensory axons, primarily from the ophthalmic branch of the trigeminal nerve, enter the cornea through the limbus and form branching subbasal plexuses that detect mechanical, thermal, and chemical stimuli, triggering blink and tear-production reflexes. In neuroscience and ophthalmology, studying this neural architecture helps explain corneal pain, sensitivity, wound healing, and nerve regeneration after injury or surgery. Imaging and functional assessments of corneal nerves also provide noninvasive indicators of peripheral neuropathy and other neurodegenerative conditions.

Functional and Morphological Assessment of Diaphragm Innervation by Phrenic Motor Neurons

Compound muscle action potential recording quantitatively assesses functional diaphragm innervation by phrenic motor neurons. Whole-mount diaphragm immunohistochemistry assesses morphological innervation at individual neuromuscular junctions. The goal of this protocol is to demonstrate how these two powerful methodologies can be used in various rodent models of spinal cord disease.

A Method to Target and Isolate Airway-innervating Sensory Neurons in Mice

Organ specific sensory neurons are difficult to identify. Fast Blue tracing is used to identify nodose neurons innervating the airways for cell sorting. Sorted nodose neurons are used to extract high quality ribonucleic acid (RNA) for sequencing. Using this protocol, gene expression of airway specific neurons is determined.

Corneal Donor Tissue Preparation for Endothelial Keratoplasty

Endothelial corneal transplantation is a surgical technique for treatment of posterior corneal diseases. Mechanical microkeratome dissection to prepare tissue results in thinner, more symmetric grafts with less endothelial cell loss and improved outcomes. Dissections can be performed at the eye bank prior to corneal transplantation surgery.
