Drosophila Brain Metabolism

Drosophila brain metabolism is the study of how the fruit fly nervous system obtains, transforms, and allocates nutrients to support neural activity, development, and survival. Neurons and glial cells process circulating sugars through glycolysis, the tricarboxylic acid cycle, and oxidative phosphorylation, while metabolic signaling coordinates energy use with nutrient availability and physiological demand. Because Drosophila offers well-defined genetics, a compact brain, and conserved cellular pathways, researchers use it to examine how metabolism influences neuronal function, aging, sleep, and neurodegenerative disease. These studies can reveal mechanisms linking disrupted energy balance to changes in brain health.

We present a protocol for measuring oxygen consumption and extracellular acidification in Drosophila melanogaster larval and adult brains. A metabolic analyzer is utilized with an adapted and optimized protocol. Micro-tissue restraints are a critical component of this protocol and were designed and created specifically for their use in this analysis.

2014

Metabolic disorders are among one of the most common diseases in humans. The genetically tractable model organism D. melanogaster can be used to identify novel genes that regulate metabolism. This paper describes a relatively simple method which allows studying the metabolic rate in flies by measuring their CO2 production.

Mitochondria play central roles in the regulation of metabolism and homeostasis. Subtle changes in mitochondrial metabolism that affect organismal physiology could be difficult to detect in whole organism metabolomics studies. Here we describe an isolation method that enhances the detection of subtle metabolic shifts in Drosophila melanogaster.
