Hypothalamus Injection

Hypothalamus injection is a stereotaxic technique used to deliver drugs, viral vectors, tracers, or other substances into defined regions of the hypothalamus, a brain structure that coordinates homeostasis, endocrine function, and motivated behaviors. Under anesthesia, researchers use a stereotaxic apparatus to guide a fine needle to target coordinates and deposit a controlled volume, limiting spread to surrounding tissue. This approach enables precise manipulation or labeling of hypothalamic circuits and supports studies of feeding, energy balance, stress, reproduction, sleep, and neuroendocrine regulation. Results can reveal how specific neuronal populations and signaling pathways influence physiology and behavior.

Despite the functional and medical importance of the hypothalamus, in utero genetic manipulation of its development has rarely been attempted. We show a detailed procedure for in utero electroporation into the mouse hypothalamus and show representative results of total and partial (regional) hypothalamic transfection.

After neural tube formation, the neuroepithelium constricts and folds while the tube fills with embryonic cerebrospinal fluid (eCSF) to form the embryonic brain ventricles. We developed this ventricle injection technique to better visualize the fluid filled space in contrast to the neuroepithelial shape in a live embryo.
