Microglia Depletion

Microglia depletion is the experimental removal or substantial reduction of microglia, the resident immune cells of the central nervous system, to clarify their roles in brain function and disease. Researchers commonly achieve it with colony-stimulating factor 1 receptor (CSF1R) inhibitors, which disrupt survival signaling, or with genetic ablation systems that selectively eliminate microglia. After depletion, microglial repopulation can be studied to examine how these cells influence neuroinflammation, synaptic remodeling, neuronal injury, and repair. In neuroscience, this approach helps distinguish microglia-dependent effects from changes caused by neurons, astrocytes, or infiltrating immune cells, supporting research into neurodegeneration, development, and brain therapies.

Microglia can influence neurons and other glia in culture by various non-cell autonomous mechanisms. Here, we present a protocol to selectively deplete microglia from primary neuronal cultures. This method has the potential to elucidate the role of microglial-neuronal interactions, with implications for neurodegenerative conditions where neuroinflammation is a hallmark feature.

Isolation of Microglia from the Cortex of an Adult Mouse Brain

The video demonstrates a technique for isolating microglia from the cortex of an adult mouse brain. First, the cortex undergoes segmentation and homogenization to remove neurons and other glial cells, preserving microglia. Following that, the cell suspension is exposed to anti-myelin magnetic microbeads and a magnetic field, effectively separating microglia and myeloid cells from myelin debris and cells containing myelin.

Macrophages play a central role in homeostasis and pathology in many tissues. The protocol presented here describes methods for depleting macrophages in vivo, deriving polarized macrophages from bone marrow aspirates, and adoptively transferring macrophages into mice. These techniques allow determination of the role that polarized macrophages play in health and disease.

In this article, a protocol for infection of macrophages with Cryptococcus neoformans is described. Also, a method for sterol depletion from the macrophages is explained. These protocols provide a guide to study fungal infections in vitro and examine the role of sterols in such infections.
