Neurosphere Culture

Neurosphere culture is an in vitro method for growing neural stem and progenitor cells as free-floating clusters, providing a model of neural development and regeneration. In serum-free medium containing mitogens such as epidermal growth factor and fibroblast growth factor, these cells proliferate and aggregate into spheres; researchers can dissociate the clusters, replate them to assess self-renewal, or withdraw mitogens to promote differentiation into neurons and glial cells. Neurosphere cultures support studies of stem cell biology, neurogenesis, disease mechanisms, and neurotoxicology, while enabling researchers to evaluate how genetic, environmental, or pharmacological factors influence neural cell proliferation and fate.

Using Primary Neurosphere Cultures to Study Primary Cilia

The primary cilium is fundamentally important in neural progenitor cell proliferation, neuronal differentiation, and adult neuronal function. Here, we describe a method to study ciliogenesis and the trafficking of signaling proteins to cilia in neural stem/progenitor cells and differentiated neurons using primary neurosphere cultures.

Here we provide a reproducible method to examine adult neurogenesis using a neurosphere assay derived from the whole brain or from either the telencephalic, tectal or cerebellar regions of the adult zebrafish brain. Additionally, we describe the procedure to manipulate gene expression in zebrafish neurospheres.
