LPSは、コアオリゴ糖である脂質AとO抗原から構成されています。

脂質Aの生合成は、ウリジン二リン酸N-アセチルグルコサミンとともに細胞質で始まり、酵素的アシル化を受けます。

これは、追加の脂肪酸が添加されるにつれて連続的に修飾されるテトラアシル化中間体を形成します。

さらなるアシル化により、ヘキサアシル化脂質Aが生成されます。次に、糖転移酵素がコアオリゴ糖を脂質Aに順次結合させます。

この脂質Aコア複合体は、MsbAトランスポーターによって内膜を横切って輸送されます。
