However, their enzymatic systems differ. Bacteria use a type II fatty acid synthase (FASII) system, where individual enzymes catalyze each step of fatty acid synthesis. In contrast, eukaryotes primarily rely on a type I fatty acid synthase (FASI), a large multifunctional protein complex that performs all reactions within a single assembly. Mitochondria, however, retain a type II-like system used for the synthesis of specific lipids such as lipoic acid.These fatty acids are ester-linked to glycerol, forming phospholipids such as phosphatidylethanolamine, phosphatidylglycerol, and cardiolipin. Membrane lipids vary across Bacteria, Archaea, and Eukarya, and these differences help cells survive in different environments. The three domains of life make lipids through distinct biosynthetic pathways. Those pathways create membranes with different stability, permeability, and flexibility.

Bacteria and eukaryotes both build fatty acid-based lipids from acetyl-CoA and malonyl-CoA. The pathway includes condensation, reduction, and dehydration steps. Bacteria use a type II fatty acid synthase, or FASII, in which separate enzymes carry out each step. Eukaryotes mainly use a type I fatty acid synthase, or FASI, which is one large complex that performs the full set of reactions. Mitochondria keep a type II-like system for making specific lipids such as lipoic acid.

These fatty acids are attached to glycerol by ester linkages to form phospholipids such as phosphatidylethanolamine, phosphatidylglycerol, and cardiolipin. These phospholipids help maintain membrane integrity, selective permeability, signaling, and protein anchoring. Because ester-linked lipids are relatively fluid, they work well in mesophilic conditions. In cold-adapted bacteria, more unsaturated fatty acids are added to keep membranes from becoming too rigid at low temperature.

Archaea use a different membrane plan. They build lipids from isoprenoid chains instead of fatty acids. These chains come from isopentenyl pyrophosphate, or IPP, and dimethylallyl pyrophosphate, or DMAPP, which are made through variants of the mevalonate pathway or other archaeal pathways. Archaeal lipids use ether linkages to glycerol, and these bonds resist hydrolysis better than ester bonds. That feature helps archaea live in harsh settings such as high heat, acid, and salt.

Most archaea form lipid bilayers, but some extremophiles, such as Thermoplasma, make glycerol dibiphytanyl glycerol tetraether, or GDGT, lipids. These lipids can span the whole membrane and form a monolayer. The monolayer adds strong stability, lowers permeability, and helps prevent membrane collapse under extreme conditions.

Some bacteria also use isoprenoid precursors to make hopanoids. In bacteria such as Bradyrhizobium and Streptomyces, squalene is cyclized to form these molecules. Hopanoids make membranes more rigid and less permeable, especially when conditions change. Their effect on fluidity is not as flexible as the effect of sterols.

Eukaryotes use the same isoprenoid precursors to make sterols such as cholesterol. Cholesterol helps control membrane fluidity by preventing membranes from becoming too rigid in cold conditions while keeping them stable at higher temperatures. This control also supports membrane protein organization and vesicle formation.

The wide range of membrane lipids shows how microbes and eukaryotes adapt to their environments. Ether-linked and ester-linked lipids, along with sterols and hopanoids, reflect evolutionary pressure on membrane structure. Lipid choice also affects membrane proteins, including transporters, enzymes, and signaling proteins. Together, these lipid changes help cells survive in many habitats, including extreme ones.
