Amino acid biosynthesis lets cells build amino acids from common metabolic intermediates. These amino acids support cell growth, protein synthesis, and metabolic regulation. The main source molecules come from glycolysis, the tricarboxylic acid (TCA) cycle, and the pentose phosphate pathway. Key precursors include alpha-ketoglutarate, pyruvate, oxaloacetate, phosphoenolpyruvate, and erythrose-4-phosphate. They supply the carbon skeletons needed to form amino acids.

Nitrogen must also be added during amino acid formation. Cells often use glutamate and glutamine as the main nitrogen donors. They help cells take in nitrogen from ammonia through the actions of glutamate dehydrogenase and glutamine synthetase. Many amino acids form through transamination, which is the transfer of an amino group to a precursor molecule. Transaminases carry out these reactions and require pyridoxal phosphate as a coenzyme.

Alpha-ketoglutarate, a TCA cycle intermediate, is the precursor for glutamate. Glutamate then acts as an amino donor in transamination reactions. It helps synthesize glutamine, proline, and arginine. Pyruvate from glycolysis is transaminated to form alanine. Oxaloacetate undergoes a similar reaction to produce aspartate.

Some amino acids need extra modification after the first steps. Aspartate can be converted into homoserine, which is an important intermediate for methionine, threonine, and lysine synthesis. Methionine biosynthesis begins when homoserine is activated to O-succinyl-homoserine. That compound is then sulfured to form homocysteine, and homocysteine is methylated to make methionine. Lysine and threonine are made through phosphorylation, reduction, and transamination steps.

Aromatic amino acids follow a separate pathway. Phenylalanine, tyrosine, and tryptophan are made from phosphoenolpyruvate and erythrose-4-phosphate. Through the shikimate pathway, these molecules combine to form chorismate, the common precursor for aromatic amino acids. Phenylalanine and tyrosine are made from prephenate. Tryptophan biosynthesis starts with anthranilate and continues through several ring-modifying steps.

Amino acid biosynthesis also affects central metabolism, so cells must replace used intermediates. They do this with anaplerotic reactions, which refill metabolic pools. Pyruvate carboxylation and phosphoenolpyruvate carboxylation restore oxaloacetate in the TCA cycle. This helps keep energy production and other biosynthetic pathways balanced while amino acids are being made.
