Glycolysis, or the Embden-Meyerhof pathway, is the first step in glucose catabolism and is universal across most organisms.This pathway consists of two stages. In the preparatory stage, glucose is phosphorylated using one ATP molecule to form glucose-6-phosphate.This intermediate is then isomerized into fructose-6-phosphate, which undergoes phosphorylation using another ATP to produce fructose-1,6-bisphosphate.Finally, fructose-1,6-bisphosphate is split into two three-carbon intermediates: glyceraldehyde-3-phosphate and dihydroxyacetone phosphate, which isomerize to yield two identical molecules of glyceraldehyde-3-phosphate.In the energy-conserving stage, each glyceraldehyde-3-phosphate molecule is oxidized to pyruvate, producing 2 NADH and 4 ATP molecules via substrate-level phosphorylation.The net energy yield of glycolysis is two ATP molecules and two NADH molecules per molecule of glucose.The final product of glycolysis is two molecules of pyruvate, which can enter aerobic or anaerobic pathways depending on cellular conditions.

The electrons harvested during glycolysis and the TCA cycle are transferred to oxygen via the electron transport chain (ETC), enabling oxidative phosphorylation. This process generates a significantly higher yield of ATP.In anaerobic conditions, alternative electron acceptors such as nitrate, sulfate, or fumarate replace oxygen in the ETC. However, many microorganisms rely solely on glycolysis for ATP production through fermentation. Glycolysis is a central pathway in glucose catabolism and a major way cells make energy. It is also called the Embden-Meyerhof pathway. This process takes place in the cytoplasm and can work with or without oxygen, which makes it useful in many organisms and conditions.

Glycolysis has ten steps and turns one glucose molecule into two pyruvate molecules. It has two phases. In the preparatory phase, the cell uses two ATP to add phosphate groups to glucose and split it into two three-carbon molecules: glyceraldehyde-3-phosphate, or G3P, and dihydroxyacetone phosphate, or DHAP. DHAP is then changed into G3P, so both molecules continue through the same later steps.

In the energy-conserving phase, G3P is oxidized, and NAD+ is reduced to NADH. This stage also uses substrate-level phosphorylation, which means ATP is made directly from high-energy intermediates. Four ATP are produced from 1,3-bisphosphoglycerate and phosphoenolpyruvate. The net result is two ATP and two NADH per glucose molecule.

Pyruvate can follow different pathways after glycolysis, depending on the organism and the environment. Under aerobic conditions, pyruvate is converted by oxidative decarboxylation into acetyl-CoA and enters the tricarboxylic acid, or TCA, cycle. The electrons collected during glycolysis and the TCA cycle move to oxygen through the electron transport chain, or ETC, which supports oxidative phosphorylation and a much higher ATP yield.

Under anaerobic conditions, other electron acceptors such as nitrate, sulfate, or fumarate can replace oxygen in the ETC. Many microorganisms, however, depend on glycolysis itself for ATP production through fermentation. In fermentation, pyruvate is reduced to lactic acid or ethanol. This also regenerates NAD+ from NADH, which keeps glycolysis running. Glycolysis is therefore a key link between cellular respiration and fermentation and helps cells maintain energy balance.
