Dimerization Binders

Dimerization binders are molecules or engineered binding agents that bring two protein partners together or stabilize their association, making protein dimerization controllable in biochemical systems. By engaging binding sites on separate molecules, these agents increase local proximity and reduce the energetic cost of forming a protein-protein interface; the resulting dimer can alter conformation, activity, or cellular localization. In biochemistry, dimerization binders support studies of cooperative binding, receptor signaling, enzyme regulation, and synthetic control of protein assemblies while providing tools for testing how molecular proximity influences function.

2014

We present a novel method of manufacturing rigid and robust short natural fiber preforms using a papermaking process. Bacterial cellulose acts simultaneously as the binder for the loose fibers and provides rigidity to the fiber preforms. These preforms can be infused with a resin to produce truly green hierarchical composites.

We present protocols for the isolation of stable heterocyclic carbenes. The synthesis of a cyclic (alkyl)(amino) carbene (CAAC) and an N-heterocyclic carbene (NHC) is demonstrated using filter cannulas and Schlenk technique. We furthermore present the synthesis of the related oxygen-sensitive, electron-rich mixed “Wanzlick dimer” and the reduced stable organic radical.

This protocol illustrates a chemically induced protein dimerization system to create condensates on chromatin. The formation of promyelocytic leukemia (PML) nuclear body on telomeres with chemical dimerizers is demonstrated. Droplet growth, dissolution, localization and composition are monitored with live cell imaging, immunofluorescence (IF) and fluorescence in situ hybridization (FISH).
