Chemical Oxygen Gradients

Chemical oxygen gradients are spatial variations in oxygen concentration that shape how cells, tissues, and engineered biological systems function. They arise when oxygen diffuses through a material or fluid while cells, microbes, or chemical reactions consume it, creating regions that range from oxygen-rich to hypoxic. In bioengineering, researchers generate and measure these gradients using controlled oxygen delivery, diffusion barriers, microfluidic devices, and oxygen-sensitive sensors. Such systems help model tissue physiology, guide tissue engineering, evaluate cell responses to hypoxia, and study oxygen transport in organ-on-chip platforms, improving the design of biomaterials, bioreactors, and therapeutic constructs.

The control of chemical and oxygen gradients is essential for cell cultures. This paper reports a polydimethylsiloxane-polycarbonate (PDMS-PC) microfluidic device capable of reliably generating combinations of chemical and oxygen gradients for cell migration studies, which can be practically utilized in biological labs without sophisticated instrumentation.

Characterization of Sickling During Controlled Automated Deoxygenation with Oxygen Gradient Ektacytometry

Here, we present oxygen gradient ektacytometry, a rapid and reproducible method to measure red blood cell deformability in samples from patients with sickle cell disease under controlled deoxygenation and reoxygenation. This technique provides a way to study red blood cell sickling and to monitor sickle cell disease treatment efficacy.

Agarose-Based Model Ecosystem for Cultivating Methanotrophs in a Methane-Oxygen Counter Gradient

A protocol is described for preparing a simple model ecosystem that recreates the methane-oxygen counter gradient found in the natural habitat of aerobic methane-oxidizing bacteria, enabling the study of their physiology in a spatially resolved context. Modifications to common biochemical assays for use with the agarose-based model ecosystem are also described.
