Dna Rna Separation

DNA/RNA separation comprises laboratory methods used to isolate DNA and RNA from one another or from proteins, lipids, and other cellular components, enabling accurate analysis of genetic material. These methods exploit differences in nucleic-acid chemistry, size, charge, solubility, and enzyme sensitivity; for example, silica-based columns bind nucleic acids in chaotropic salts, while selective DNase or RNase treatment removes unwanted DNA or RNA. Electrophoresis and chromatography can further separate molecules by size or interactions with a stationary phase. Purified DNA and RNA support PCR, sequencing, gene-expression studies, diagnostics, and investigations of cellular structure and function.

Separation of Single-stranded DNA, Double-stranded DNA and RNA from an Environmental Viral Community Using Hydroxyapatite Chromatography

We describe an efficient method to separate single-stranded DNA, double-stranded DNA and RNA molecules from environmental viral communities. Nucleic acids are fractionated using hydroxyapatite chromatography with increasing concentrations of phosphate-containing buffers. This method permits the isolation of all viral nucleic acid types from environmental samples.

Here, we present a protocol for tracing genomic DNA (gDNA) contamination in RNA samples. The presented method utilizes primers specific for the internal transcribed spacer region (ITS) of ribosomal DNA (rDNA) genes. The method is suited for reliable and sensitive detection of DNA contamination in most eukaryotes and prokaryotes.
